Comparison of argyrophil nucleolar organizer region counts, proliferating cell nuclear antigen (PCNA) indices and mitotic indices in fibromas and fibrosarcomas

Eighteen fibromas and four fibrosarcomas from various species were studied using three different methods of assessing aspects of cellular proliferation; these were immunostaining for proliferating cell nuclear antigen (PCNA) clone PC10, interphase nucleolar organizer region staining (AgNORs) and mitotic indices. The tumours were from 22 different animal species, breeds and ages, between the 1980s and 1995. All samples were fixed in formalin and embedded in parrafin wax. Nucleolar organizer regions associated with argyrophil proteins were evaluated quantitatively on specific silver stained cells. The mean number of AgNORs was determined in at least 100 cells of each lesion. PCNA-labelled fractions and mitotic indices were estimated in 10 high-power fields. Both AgNOR counts (P0.01) and PCNA indices (P0.05) were significant in the Mann-Whitney U test, but mitotic indices were not significant. Thus, only AgNOR counts and PCNA indices can separate benign from malignant lesions and can be considered a statistically significant tool in the evaluation of fibromas and fibrosarcomas
