Starch accumulation as a marker for microtuberization in potato (Solanum tuberosum)

The first sign of starch accumulation in potato axillary buds as an indicator of microtuber induction and initiation under different photoperiod and temperature conditions was studied. Single-node potato (Solanum tuberosum cv. Bintje) explants were inoculated on tuberization medium (MS (Murashige and Skoog (1962)) with 8 % sucrose and 22 micro M BA (N sup(6) benzyladenine)) and on MS medium with 8 % sucrose (control medium). Cultures were incubated at 20 deg C in short day conditions and in darkness, and 26 deg C in long day conditions and in darkness. Starch deposition occurred on day 3 in basal part of axillary buds cultured on tuberization medium at 20 deg C and 12 hrs photoperiod starch began to accumulate on day 8, and in darkness on day 12. In explants grown under the same conditions on control medium starch deposition was not observed after two weeks. A higher temperature delayed initiation and development of microtubers, especially in darkness, despite whether or not the tuber-inducing medium was used.
