Assay of aflatoxin producing fungi by culture filtrate spot method

Fungi isolated from corn and peanut were examined for aflatoxin production. The toxin was assayed by the culture filtrate spot method. Isolated fungi were cultured on YES medium and incubated for 7 days at room temperature. The culture filtrates were subjected to TLC plate for aflatoxin detection. The TLC plate was developed in the chloroform, acetone and N-Hexane (85:15:20) mixture for 30 minutes. Aflatoxin was viewed under UV lamp (365 nm) in the presence of standard aflatoxin.
