The karyotype of Distichodus affinis was analyzed by means of sequential banding: Giemsa-, C-banding, and silver-staining and CMA3 fluorescence staining. The diploid chromosome number was 2n = 48 (16 pairs of metacentric and 8 pairs of submetacentric chromosomes). The nucleolar organizer regions (NORs) were situated in the pericentromeric region of one medium-sized metacentric chromosome pair. The karyotype contained very low content of C-positive heterochromatin. No heteromorphic sex chromosomes were detected. This first report for any of the African characiform fishes stresses the need of other karyotypic data to reveal cytotaxonomic relationships of both African and South American characiform fishes.
