Vitamin B12 production for food and feed using local isolates and agro-industrial by-products as substrates: terminal report

Three Biotech strains, all belonging to the Bacillus genus namely, Bacillus 1301, 1303, and 1304 were evaluated for vitamin B12 production using coconut water plus salts as medium. In comparison with standard Vitamin B12 producers, Pseudomonas denitrificans ATCC 13687 and Propionibacterium shermanii ATCC 13673, Bacillus 1301 exhibited the best producing ability for B12 production. The optimization of fermentation conditions was carried out on laboratory scale using a 2.6 L fermentor equipped with temperature and pH controls. Fermentation conditions such as sugar concentration, aeration and agitation were studied. Three fermentation modes were carried out which are the batch, fed-batch and continuous modes of fermentation. The best mode of fermentation in this study was the standard batch and the optimum sugar concentration was 40 g/l while agitation and aeration rates were 350 rpm and 0.5 vvm, respectively. Between coconut water plus salts medium and molasses, coconut water gave the best results. There was no B12 production with the molasses runs which could only be because of the inhibitory effect of molasses on the strain used for the production. Downstream processing of the crude vitamin B12 gave a value of 5 mg/g of dried biomass. Purification studies for the production of crystalline vitamin B12 have also been started.
