Effect of plant growth regulator on the multiplication of Kerk-lily in vitro

English. Tissue culture of Lilium longiflorum have an important role on the mass production of seedling in relatively short time, and virus free. Several factors affected the successful bud and bulblet formation. This research aimed to know the effect of plant growth regulator on the multiplication of Kerk lily in vitro. Research had been conducted in the Tissue Culture Laboratory of Indonesian Coffee and Cocoa Research Institute. The research consist of two experiments i.e multiplication stage and rooting stage. For the multiplication stage, treatments was arranged factorially, i.e. IAA and BAP concentration. Three levels of IAA concentration namely 0.00; 0.15; and 0.30 mg/l, and five levels of BAP concentration namely 0.00; 2.50; 5.00; 7.50; and 10.00 mg/l. The treatments during rooting stage were NAA concentration consisted of eight levels namely 0.00; 0.15; 0.30; 0.45; 0.60; 0.75; 0.90; and 1.05 mg/l. The result showed that BAP affected bud growth, however BAP inhibited bulblet growth. The best bud growth occur on the medium contained BAP 10 mg/l with the number of maximum bud was 8.2 per explant. The maximum number of bulblet from one explant was 9.1 from the treatment of IAA 0.15 mg/l. NAA was an effective auxin for rooting induction of lily micro shoot. The best percentage of rooting micro shoot was 82.03 percent from the treatment of NAA 0.79 mg/l

Kultur jaringan pada tanaman Lili (Lilium longiflorum) mempunyai peranan yang penting untuk menghasilkan tanaman dalam jumlah besar dalam waktu yang singkat terutama untuk memproduksi bibit kultivar baru secara cepat atau tanaman yang bebas virus. Dalam teknik kultur jaringan lili terdapat beberapa faktor yang berpengaruh terhadap keberhasilan pembentukan tunas maupun bulblet. Penelitian ini bertujuan untuk mengetahui pengaruh zat pengatur tumbuh terhadap perbanyakan Kerk lili secara in vitro. Penelitian dilaksanakan di Laboratorium Kultur Jaringan Pusat Penelitian Kopi dan Kakao Jember. Untuk proses perbanyakan, perlakuan disusun secara faktorial, yaitu faktor konsentrasi IAA dan BAP. Faktor IAA terdiri atas 3 aras yaitu 0,00; 0,15; dan 0,30 mg/l. Faktor BAP terdiri atas 5 aras yaitu 0,00; 2,50; 5,00; 7,50; dan 10,00 mg/l. Untuk proses pengakaran perlakuan berupa konsentrasi NAA yang terdiri atas 8 aras yaitu 0,00; 0,15; 0,30; 0,45; 0,60; 0,75; 0,90; dan 1,05 mg/l. Hasil penelitian menunjukkan bahwa BAP berpengaruh terhadap pembentukan tunas akan tetapi bersifat menghambat pembentukan bulblet. Pertumbuhan tunas terbaik terjadi pada media yang mengandung BAP 10 mg/l dengan jumlah tunas per eksplan maksimum 8,2. Jumlah bulblet maksimum yang dihasilkan per eksplan adalah 9,1 diperoleh dari IAA 0,15 mg/l. NAA merupakan auksin yang cukup efektif untuk menginduksi akar adventif tunas mikro lili. Persentase tunas mikro berakar terbaik sebesar 82,03 persen diperoleh dari perlakuan NAA 0,79 mg/l
