Callus formation and plantlet regeneration on different tissue explants of chrysanthemum

Different varieties of chrysanthemum were evaluated for callus formation and organogenesis using such tissue explants as leaf strips, petioles, nodes and internodes from aseptic stock cultures and petals of field-grown plants. These were cultured on Murashige-Skoog (MS) medium supplemented with varying levels of NAA applied singly or in combination with BAP. 2,4-D was also used on petals. Callus formation and organogenesis varied with genotype, explant tissue and level of combination of growth regulators. Taiwan Yellow readily formed callus and regenerated shoots on all explant types over a range of treatments but more shoots were formed on leaf strips that formed callus cultured on MS medium supplemented with 0.6 mg/l NAA and 0.4 mg/l BAP. Origin of regenerant, whether from callus or explant tissue, was however, not established
