Influence of the black elder (Sambucus nigra) pomace extract on hypercholesterolemia, lipoperoxidation and carcinogenesis of rat colon

The influence of the diet supplemented by 4% of the extract form black elder (Sambucus nigra) pomace on the colon carcinogenesis induced by dimethylhydrazine (DMH) was examined in just weaned male Wistar rats fed diet containing 0.3% cholesterol. DMH was administered subcutaneously once a week at a dose of 20 mg/kg either three times (8 week experimet) or twelve times (28 week experiment). At the end of the experiments, the animals were sacrified after a 18-h food removal by decapitation at a light ether anesthesis. The extract of black elder did not influence hypercholesterolemia either in the shorter or in the longer experiment. In the longer one, a decrease in the cholesterol content in liver and, oppositely, an increase in the serum triacylglycerol level was observed. In both experiments, the diet supplemented with the black elder extract lowered the content of conjugated dienes in colon by as much as 40% and, in the longer one, the content of conjugated dienes significantly decreased also in liver. In the longer experiment, the diet supplemented with black elder extract decreased the activity of superoxide dismutase, glutathione peroxidase and glutathione transferase in liver. In the longer experiment, the diet supplemented with the black elder lowered the number of moderate and large aberrant crypts foci by 54-80% and their total number by 16%. The black elder extract-induced decrease in the incidence and volume of tumors was not significant because of a great variance of the data
