Multiplication of teak seedlings from genetic proved seeds through tissue culture techniques

Multiplication of teak seedlings were studied by taking genetic proved seeds from Mae May clonal seed orchard at Lampang province to the Faculty of Forestry, Kasetsart University for furture experiments. The seeds were germinated to be seedlings on modified MS and White media. The shoot, nodal and hypocotyl with cotyledon explants from seedlings were induced shoots and roots on MS media supplemented with plant growth regulators. Rooted shoots or plantlets were transplanted to planting media and determined survival percentages. Teak seeds germinated and developed to be healthy seedlings with the greatest average number of nodes, 2.5 nodes, when cultured on 1/4 strength of the modified MS medium for 30 days. Media, subculture cycles and genetic of the clones were the major factor affecting multiplication of the shoots and buds. The MS medium supplemented with BAP 2 mg/l was the most effective medium for multiplying shoots and buds up to 2.07 shoots/explant at 30 days subculture interval. The shoots obtained could be induced roots on the MS medium either with or without auxin. NAA was more effective auxin than IBA in stimulating roots. The MS medium supplemented with 0.5 mg/l of NAA induced good quality roots of in vitro teak shoots. Increasing the number of subculture cycles tended to reduce rooting percentage and the number of roots per one shoot. The survival rate of transplanted teak plantlets was 97-100 percent after 30 days when planted in rice husk charcoal under at high air humidity.
