Tissue culture for the propagation and development of papaya that are tolerant to papaya ringspot virus

Development of PRV-tolerant papaya was carried out by crossing tolerant variety (Florida) to local susceptable variety (Kackdum). F1 hybrids were selected under field condition. The selected ones were tolerant to PRV and had good fruit production. Axillary buds from the selected plants were excised and aseptically cultured in the appropriate medium for multiplication. Papaya plants in vitro showed the symptom of virus infection. Therefore, virus elimination through shoot tip culture was done and detected by Elisa Test. The virus free plants were propagated for mass production. The complete plants were transferred to soil. The plants derived from tissue culture were grown in the field. They showed good fruit production and tolerant to PRV. The production of the same clone had the same uniform fruit characteristics. From the experiment, somatic embryos were also induced from root callus culture. Mass production of the embryos could be carried out and after that being regenerated to seedling. The mature seedling were successfully transferred to soil. This technique is a very useful tool for improving new varieties of papaya in the future.
