Diagnostic aspects of bovine spongiform encephalopathy are presented. They are based on the epizootological data, clinical symptoms, the post mortem analysis of the CNS including inoculation of adapted recipients and results of the histopathology, demonstration of the specific scrapie-associated fibrils by negative-stain electron microscopy in detergent extracts of fresh brain tissues, immunohistochemical detection of the abnormal deposits of PrPsc in formalin fixed brain tissues, the detection of the constituent PrPsc by electrophoretic separation and immunoblotting techniques as well as the PLATELIAtm BSE test manufactured by Bio-Rad Laboratories which is based on the sandwich immunoassy technique, an adapted ELISA test designed to be fast, sensitive and appropriate for mass screening.
