Ultrasound-guided follicular aspiration and oocytes collection in reproductive disturbed cows

Ultrasound-guided follicular aspiration was employed to evaluate the possibility of collection of oocytes and production of embryos in a normal cycling cow (A) and 4 cycling cows that had been some disturbed of reproduction, that is endometritis (B), silent ovulation (C), disturbance of estrous cycle (D) and hypoovaria (E). The objective of first experiment was to examine the change of number of follicles on ovary by aspirating the follicular fluids repeatedly. The monitoring of follicles was replicated four times every 3 or 4 days before follicular aspiration was started. In order to determine the effects of follicular aspiration on the ovarian activity, the second experiment was designed to measure the plasma progesterone concentrations. In both experiments, a 5MHz linear-transducer with needle connected to an ultrasound scanner was used for the detection of follicular dynamics and the follicular aspiration every 3-4 days. Follicular fluids were aspirated from all follicles in diameter more than 4 mm. The morphology of oocytes or oocytes-cumulus complexes collected from disturbed 3 cows was very similar and poor quality. The proportion of denuded or partially denuded oocytes as a fraction of the total oocytes collected by transvaginal aspiration was higher than that of oocytes collected directly from ovaries harvested at slaughter. Oocytes with normal morphology collected from normal cow were fertilized and cultured in vitro. Of oocytes cultured from G1 randk 38%, G2 rank 24% and G3 rank 6% developed to blastocysts. None of them from G4, G5 and G6 rank developed to blastocysts
