Viability and growth rate of gram positive, sporing and gram negative, non-sporing bacteria: A compact contemplation

Bacillus subtilis, a gram positive (g+ve) sporing, and Escherichia coli, a gram negative (g-ve) non-sporing bacteria, were used as model of present study and were tested for their viability and growth rate at different centrifugation speeds (1000-5000 rpm) for 10, 20 and 30 minutes. The centrifuged cells were also tested on different agitation speeds of rotary shaker incubator at 50, 80, 110, 140 and 170 rpm for 24 hours at 37 (degree centigrade) at UV absorption 675 nm. The present study revealed the viability and highest growth rate i.e. Optical Density (O.D.) of both bacteria at 4000 rpm. when inocula were centrifuged for 20 minutes in centrifuge machine and at 110 rpm in shaker incubator at 37 (degree centigrade) for 24 hrs. post-incubation in nutrient broth supplemented with 1% weight/volume (w/v) glucose.
