Green fluorescent protein as a tool for in vivo examination of bacterial cells with the use of confocal microscopy

The in vivo study of bacterial cells was often limited because of the few adequate detection methods and modes of genetic analysis. The green fluorescent protein, GFP, originally found in the jellyfish Aequorea victoria, can also be used for genetic analysis of the prokaryotic cells. Even if there is only one copy of the gfp gene in the bacterial chromosome, the GFP fluorescence is usually high enough. For in vivo examination of eukaryotic cells confocal microscopy was found adequate for the examination of cells in real time and space. It is also possible to apply confocal microscopy for examination of prokaryotic cells. GFP can be used for the development of different reporter systems. The present article describes promotor analysis construction of different transcription fusions. Such reporter systems are available (i) on a plasmid, (ii) on minitransposon Tn5, located on a suicide plasmid.
