Number and quality of buffalo oocytes recovered relative to method of harvest, stage of estrous cycle and corpus luteum

Number and morphology of oocytes harvested from ovaries were correlated to 3 methods of oocyte collection, viz. aspiration, individual follicle isolation (IFI), and whole ovary slicing (WOS), stage of estrous cycle and presence or absence of corpus luteum. Paired ovaries were obtained from non-pregnant buffaloes at slaughter and stage of cycle was estimated by visual examination of the corpus luteum. The IFI method yielded highest number of total and culturable oocytes per pair of ovaries. The average mean number of culturable oocytes identified on the basis of gross morphology in aspiration method (0.26/ pair) were significantly lower than IFI (0.88/ pair) and WOS (0.68/ pair) methods. However, no significant difference was observed between IFI and WOS methods. Also no significant difference was observed between number of total and culturable oocytes collected from ovary at different stages of cycle. Ovaries with CL yielded significantly higher number of culturable oocytes per ovary than ovaries without CL, suggesting that CL could exert its effect by decreasing atresia.
