Genetic engineering at the Philippine Rice Research Institute (PhilRice) supports the existing strategies to improve pest resistance in the high yielding varieties, new plant type elite lines and the B lines used in hybrid breeding. Plant growth regulators, genotype, explant type, and tissue culture conditions were optimized for Agrobacterium tumefaciens-mediated transformation. Low transient expression efficiencies in the inbreds and hybrid parentals were obtained from calli derived from mature seeds, while higher efficiencies were obtained from calli derived from immature embryos and young inflorescence after co-cultivation with LBA4404 (pTOK233). The new plant type lines containing the japonica background showed high transformation efficiencies. Transgenic plants containing pest resistance genes for bacterial blight, blast, sheath blight, and stemborer resistance are being developed using the optimized conditions in the different genotypes. Currently, putatively transformed transgenic plants of PSB Rc14, 28, IR72, IR64, B lines 913 B, and PR4B containing the chitinase and glucanase genes were obtained.
