Biochemical and molecular-based characterization of pathogenic and nonpathogenic strains of marine Vibrio

Certain memebers of the marine Vibrio species have been implicated in shrimp mortality in Philippine aquaculture farms. In this study, ten marine Vibrio strains were obtained from several Philippine shrimp farms. Initial pathogenecity testing of these isolates revealed that five of these were pathogenic to black tiger shrimp (Penaeus monodon), while the other five were classified as nonpathogenic. Further characterization of all ten isolates were performed using standard phenotypic tests, PCR of randomly amplified polymorphic DNA (RAPD-PCR), and denaturing polyacrylamide gel electrophoresis (SDS-PAGE). Differences among the ten strains were observed using combined phenotypic-and molecular-based approaches. Characteristic phenotypes and molecular based profiles were also noted for both the pathogenic and the nonpathogenic subgroups. Distinct differences in RAPD profiles can possibly pave the way for developing DNA based protocols to detecting members of marine Vibrio commonly isolated from shrimps farms. One of these isolates have been identified to belong to Vibrio campbelli while the rest were also observed to be very closely related to the species based on partial 16S rDNA sequences.
