[Isolation and cultivation of ectomycorrhizal fungus mycelium]

Isolation and manipulation of ectomycorrhizal fungus cultures are crucial phases in artificial mycorrhization of forest tree seedlings. Isolation, cultivation and maintenance procedures of pure cultures of ectomyorrhizal fungi are described and discussed. Pure cultures are isolated from sporocarp tissues to test tubes on agar nutrient media, cultivated in dark at room temperature and stored within subcultivation cycles approximately 3 months under refrigeration until the next transfer period. BAF and MMN nutrient media are preferred for isolation and manipulation of cultures in our laboratory. Higher rates of agar cultures are cultivated in Petri dishes and submerged cultures in Erlenmeyer flasks. At present, pure cultures of 21 species (33 isolates) of obligatory ectomycorrhizal fungi are available in Department of Silviculture of Technical University in Zvolen for preparation of fungal vegetative inocula
