Histological and glycoconjugates histochemical studies on skin glands of the buffaloes

In the muzzle skin glands of the buffaloes, histology and glycoconjugates histochemistry have been studied by means of conventional and lectin histochemical procedures. The conventional staining methods employed were Hematoxylin and Eosin (H&E), Masson Tricheme, Alcian blue pH 1.0, Alcian blue pH 2.5, Low iron diamine (LID), High iron diamine (HID), AB pH 2.5-PAS and HID - AB pH 2.5. In addition 7 lectins, Concanavalin A (Con A), Ricinus communis agglutinin - I (RCA-I), Peanut agglutinin (PNA), wheat germ agglutinin (WGA), Ulex europeus agglutinin-I (UEA-I), Limax flavus agglutinin (LFA), and Dolichus biflorus agglutinin (DBA) were applied to detect saccharides residues. According to the results obtained, epidermis of the muzzle skin is thick and consists of stratum corneum, stratum granulosum, stratum spinosum and stratum germinativum. The number of melanin granules decreases as cells move to the surface of the epithelium. Aggregates of labial glands are distributed in the propria-submucosa. The secretory endpieces found to be seromucous type. The seremucous cells stained strongly to moderately positive with AB pH 2.5, LID, PAS, Con A; weakly positive with RCA-I, WGA, DBA and LFA. And negatively with UEA-I and PNA. In contrast, the intercalated duct cells found to exhibit strong reaction with WGA, UEA-I, DBA and PNA. This finding suggests that the secretory glycoconjugates are various types in chemical nature and their physiological cativities.
