Production of Amorphophallus for agar industry: Testing method of Amorphophallus gel

Glucomannan flour had been produced from the corns of elephant foot yam (Amorphophallus oncophyllus). The overall properties of the flour produced at 3 different methods showed some inconsistency in their purity and turbidity comparing to normal gelatin for the same concentration. However, the mixing ratio of glucomannan: gelatin at 6:2 to make the concentrated gel of 8 gm/litre gave comparatively the same turbidity of using just gelatin. As for the use of this mixture of glucomannan gel in synthetic culture medium VW and MS, for 24 kinds of orchid seed germination and 4 kinds of orchid tissue culture, as well as 13 other plants, it was found that glucomannan was quite superior in producing higher quantity of plants, inducing more shoot tips, leaves, and roots than using the gelatin alone. As for growing bacteria Erwinia carotovora and Fusarium oxysporum, it was also found that using glucomannan in the medium was promising but deep brown liquid was occured in bacteria culture while glucomannan medium for fungi was not changed to violet.
