Identification of Rhizoctonia solani from Belgian sugar beet growing area

Rhizoctonia solani Kuehn [teleomorph: Thanatephorus cucumeris (Frank) Donk] is a complex fungal species with varying ecological, morphological, physiological and pathogenic properties. Hitherto, Rhizoctonia solani has been subdivided in so-called anastomosis groups (AGs). However, AG-system gives little information neither about the genetic variability within each AG nor about the taxonomic relationships between the different AGs. For these reasons, there is an increasing interest in novel approaches based on biochemical, serological and molecular techniques. An attempt was made to characterize Rhizoctonia solani strains isolated from infested Belgian sugar beet fields. 37 isolates obtained from different locations were divided into groups using microscopic study of the anastomosis, and analyses of polymorphism of the pectic enzymes, polymorphism of rDNA and differences in total protein composition. It was concluded that all isolates belong to AG 2-2 3B. Analysis of pectic zymograms allowed classifying most of the isolates into two homogeneous groups. Three isolates of different origin assessed on sugar beet seedlings and on 14 weeks old plants of the very same AG exhibit clear differences in pathogenic properties.
