Specific middle piece staining of sperms and possible applications

A special staining technique for the middle piece of sperms is described in different species (cow, horse, sheep, goat, pig). Staining was performed with frozen or fresh semen. The mitochondria in the middle piece could not always be stained with the same intensity. Using 600-fold magnification, subunits of the middel-piece could be observed, but only on fresh collected and immediately stained sperms. When kept in the refrigerator for more than 3 days, the observed subunits disappeared. A positive correlation between staining intensity and enzyme activity could be indicative for sperm motility and also fertilizing capacity.
