Genetic diversity analysis of tall coconut from Mollucas employing random amplified polymorphic DNA

Indonesia represents a large potential genetic resources for genetic improvement of coconut palms (Cocos nucifera L.). This study was carried out in March-October 1999 by means of RAPD analysis on a representative coconut leaf samples from Mollucas grown as ex situ collection in Mapanget, Manado i.e. Takome Tall (DTE), Igo Bulan Tall (DIB) and Igo Duku Tall (DID) populations. The objective of this research was to study the genetic diversity of Tall coconut populations from Mollucas based on RAPD markers. Twelve random primer were used to amplify DNA employing Polymerase Chain Reaction (PCR). Five primers generated 29 polymorphic DNA fragments. The grouping of the individual coconut tree according to the cluster analysis was in good agreement with their fruit shape. Genetic distance within DTE, DIB and DID populations was 8, 3 and 6 percent respectively, while among populations was 20 percent. The low of the genetic of those populations might be due to self or cross pollinated within each coconut population in the fields, OPA-13 primer revealed 600 bp DNA fragment found only in the DIB population while a 300 bp DNA fragment found in the DID population
