[Kinetics of hydrolysis of carbohydrate and protein on solid substrate fermentation with Rhizopus oryzae]

Solid substrate fermentation of tempe has been known for centuries in Indonesia. The technology of this fermentation is used for conversion of substrate into edible food. The use of this technology for bioactive compound production, however, is limited due to the low hydrolysis of substrate by the molds. This experiment is conducted to study the kinetics of hydrolysis of carbohydrate and protein of leucaena seeds and winged beans as substrate fermented with Rhizopus oryzae for 3 days of fermentation. Results showed that during fermentation substrate of both beans was decreased. The total loss of the total solid was 2.3 g for leucaena seed and 1.2 g for winged beans. There are increased in soluble sugar and amino-N in the substrate. The pH and acidity was also increased during fermentation. The increased of soluble sugars for 3 days of fermentation was 20 times in leucaena seed and 29 times in winged beans. The kinetics data of hydrolysis of the carbohydrate and protein were fit with the Michaelis-Menten equation for substrate concentration and rate of production of sugar and amino-N relationship. The micronmax of leucaena seeds fermentation were 0.1190 g/hour and 0.0039 g/hour for sugar and amino-N respectively. For winged bean substrate the micronmax soluble sugar and amino-N were 0,0640 g/h and 0,0036 g/h, respectively. There is a relationship between rate of hydrolysis and both total solid loss and surface area of the substrate. The solid substrate fermentation for production of specific product such as bioactive compounds should consider the relationship between spore concentration with surface area of substrate and the hifa penetration of the mold and these data could be used to develop the bioreactor specifics for these purposes
