Description and in vitro regeneration of three citrus rootstocks

Three citrus rootstocks Bitter Sweet orange, yuma citranges and sour orange were described for their morphology, fruits and seeds. Regeneration response of these rootstocks tinder in vitro was also studied. Germination percentage of peeled seeds was higher in yuma citranges (73.33 %). Callus induction % age was also higher in leaf explants of yuma rather node, inter-node and cotyledons on MS media under, light Under dark condition internodes showed good performance while cotyledons produced no callus in these rootstocks. On MS media leaf explants had yielded maximum callus growth followed by internodes. Bitter Sweet orange showed the best performance because this changed its colour completely from light brown to green under dark conditions while under light condition maximum callus growth was in deaf explants. On MT media under light conditions, leaf explants had the highest callus growth. Sour orange callus changed its colour from brown to light brown. Using the same media under dark condition leaves and internodes, showed good callus growth. Morphological studies describe that tree shape of bitter sweet orange was spheroid with dense branches, spreading growth habit, wide branch angle while spines were present in the interior side were of straight shape. Leaves were simple with dark green colour, medium size and brevipetiolate. Better sweet orange yielded the maximum fruit weight (195.96 g). Fruit skin was yellow with rough surface adherence of albedo was strong while some had medium adherence. Seed were of clavate shape few were of spheroid shape. It has the highest number of seeds. In yuma citranges tree shape was spheroid with medium branch density and spreading growth habit. Fruits was the highest of all with small size. Fruit had spheroid shape with convex base, truncate apex and yellow skin, rough surface. Juice content was tow and vesicles were short, seeds were monoembryonic. Sour orange had ellipsoid tree shape, dense spreading branches, wide branch angle, less number of spines were present on exterior and more on interior of tree and green shoot tip colour. Fruit weight, diameter and fruit length was medium. Ellipsoid fruit shape was prominent, few shape was prominent, were of spheroid. Fruits showed convex base, truncate apex and yellow colour. Surface texture was rough and medium adherence of albedo with white colour. Seeds were of clavate shape, medium in number, smooth surface.
