Renal clearance of acetylsalicylic acid in male volunteers after its oral administration

Renal clearance of endogenous creatinine and salicylic acid was investigated in twelve healthy human male volunteers after oral dose of (2x300 mg) tablets of aspirin (acetylsalicylic acid). Blood and urine samples were collected at pre-determined time intervals and analyzed for creatinine and salicylic acid concentration by calorimetric method. The average plus minus SE for the rate of urine flow was 0.031 plus minus 0.011 ml/min/kg; pH of blood was 7.67 plus minus 0.018 and pH of urine was 6.17 plus minus 0.059. Concentration of creatinine in plasma and urine was 15.11 plus minus 0.166 and 855.73 plus minus 1.52 mug/ml respectively. The mean plus minus SE value of salicylic acid in plasma and urine was 29.378 plus minus 0.118 and 38.303 plus minus 0.427 mug/ml respectively. Renal clearance ratio was 0.054 plus minus 0.016. The studies indicate that the renal clearance of endogenous creatinine in male volunteer in according to the value i.e., 10-20 ml/min/kg. A non- significant positive correlation between dieresis and renal clearance of salicylic acid shows back diffusion or re- absorption. There is significant positive correlation between urine pH and salicylic acid clearance. So, pH of urine has significant influence on the renal clearance of drug. There is non- significant positive correlation between dieresis and clearance ratio and clearance ratio. There are non-significant negative correlation between plasma concentration and clearance ratio and plasma concentration and drug clearance as shown by results and graphs. It is concluded that besides glomerella filtration, mechanism of back diffusion or re- absorption was also involved in salicylic acid reaction.
