Detection of newcastle disease virus in the intestinal contents of broilers and layers

Two hundred intestines samples (hundred each of broilers and layers) of about 8 cm were collected from the poultry sale shops of Faisalabad city. These pieces were opened scratched and vigorously shaken into the sterilized normal saline and the suspension was centrifuged. The supernatants were subjected to spot haemagglutination with 2% chicken RBC's. Out of total samples 95% samples of layers and 75% of the broilers showed positive spot haemagglutination activity. On micro Haemagglutination inhibition with known ND antiserum 85 samples and 66 samples, were positive in layers and broilers respectively. 10% samples of the layers and 9% of the broilers were not inhibited by specific Newcastle disease antiserum suggesting other HA viruses such as avian influenza and infectious laryngotracheitis viruses. A total of twenty samples ` were isolated in embryonated eggs. (Chorio-allantoic route). These isolates were confirmed as NDV by means of haemagglutination inhibition test. The pathotyping of the five isolates was conducted on the basis of intracerebral pathogenicity index (ICPI) in day, old chicks. Values obtained were 0.28, 0.31, 0.37, 0.38 and 0.46 from each isolates. The isolates were found to be related to the lentogenic strains of healthy broilers, and layers is suggestive of mutation consequently outbreaks of Newcastle disease among broilers and layers.
