Urinary excretion of trimethoprim in female volunteers

Trimethoprim is an antimicrobial drug used for the treatment of infections of upper and lower respiratory tract, tract and middle ear. Studies on the excretion of trimethoprim yield valuable knowledge for the rational used of drug. The trimethoprim tablet of 164 mg was given orally to healthy female volunteers and its excretion in urine was monitored. Urine samples were collected at pre determined time interval after the drug administration. The results showed the average 4 SE values for diuresis, pH and rate of excretion of trimethoprim were 0.019 plus minus 0.004 mL/min/kg, 6.56 plus minus 0.155 and 0.117 plus minus 0.018 mug/min/kg respectively. The average plus minus SE values for concentration of trimethoprim in urine of female volunteers at 60, 20, 180,240, 360, 480 and 720 minutes as mg dose excreted in urine was 0.843 plus minus 0.107 0.999 plus minus 0.114, 1.24 plus minus 0.144, 1.42 plus minus 0.182, 1.52 plus minus 0.173, 1.78 plus minus 0.301 acid 2.23 plus minus 0.370 mg respectively. The results showed the average plus minus SE values cumulative dose was 4.862 plus minus 1.153 mg. Percent cumulative dose excreted in urine was 6.26%. Trimethoprim is basic drug which is excreted move in acidic urine. The excretion of trimethoprim is 6.26 percent during 12 hours. This differences may be due to hot weather, by volunteers, and acidic pH of urine. The study supports the need for comprehensive evaluation of drug under local environment to obtain pharamacokinetic parameters on which the rational dose regimens of drugs could be based.
