A study of alpha-amylase production by Bacillus subtilis through solid state fermentation

The main objective was to study effects of corn steep liquor, yeast extract, SDS and Tween-80 by using pre-optimized basal medium. Kinetic studies of enzyme were also included in the study to characterize the cc- amylase produced by Bacillus subtilis when banana peels were used as substrate in SSF. Pre-optimized basal medium was used with standard culture condition for optimized growth and cc-amylase production as earliest. It was observed that Bacillus subtilis produced maximum (x-amylase in the a growth medium containing (0.1%) yeast extract, 1% corn steep liquor, 0.15% sodium dodecyl sulphate and 0.1% Tween-80 under pre- optimized culture conditions. The alpha- amylase produced under optimum conditions was characterized by studying the effect of varying conditions of pH and temperature separately and in various conditions. Stability of enzyme at different temperature and after specific time periods was also studied, alpha-amylase showed maximum activity at pH 6 and 60 degree C. alpha-amylase retained 84.29% of its activity upto hours at optimum pH and temperature. The pH and temperature optimum characteristics gave an indication that the enzyme is suitable for industrial applications after complete characterization.
