Excretion of trimethoprim through urine of male volunteers

Many factors can influence individual's response to a drug including age, disease of organs. of drug elimination (kidney liver), use of other drugs, food and chemicals, genetic factor and environmental conditions. It is likely that the genetic and environmental bases of drug reaction can be discovered and screening techniques can be applied to assess individual's risk. Septran tablet (trimethoprim 160 mg + sulphamethoxazole 800 mg) was given to each volunteer: orally and urine samples were collected at 60, 120, 180, 240, 360, 480, 720 minutes after drug administration. The samples were analyzed by spectrophotometer. The results show that the average plus- minus values for diuresis was 0:026 plus-minus 0:007 mL/min/kg, for pH of urine excreted was 6.44 plus-minus 0.164, for the concentration of trimethoprim excreted in urine of male was 11.676 plus-minus 2.77 (ug/mL). The average rate of excretion was 0.185 plus-minus 0.04 ug/min/kg. Average amount of drug excreted was 2.143 plus-minus 0.432 mg, the percentage of the drug excreted was 1.307 plus- minus 0.272% The mean plus-minus SE values for the cumulative dose excreted in the urine of male volunteers was 8.040 plus-minus 11.211 ug and cumulative percentage dose excreted was 5.02 plus-minus 0.755 %. There is. significant negative co-relation between rate of excretion and pH of urine. The trimethoprim is basic drug and excreted in acidic urine. The excretion of trimethoprim was 9.667 % during twelve hours. In another study the excretion of trimethoprim was 6.259 plus-minus 2.140% (Munir 2002). The differences may be due to variability in gender, fluctuation in urine pH and nutritional ingredients.
