DNA preparation for seahorse genetics analysis

Total DNA of seahorse Hippocampus histrix was extracted from whole blood preserved in TNES-Urea buffer (10 mM Tris-HCl, pH 7.5; 125 mM NaCl; 10 mM EDTA; 1 percent SDS; Urea 4 M). The DNA yield was about 2-5 micro g of total DNA/micro l blood. The molecular mass of the obtained DNA was high with greater than 23.1 kbp in length. After digested with BamHI, EcoRI, PstI, and HaeIII and amplified at 18S rRNA gene, this indicated that the total DNA was suitable for further genetic analysis.
