Production of antibody against vitellogenin for gonad inhibiting hormone assay in Metapenaeus affinis

Antiserum against vitellogenin of the shrimp Metapenaeus affinis was produced by immunizing the first set of mice with shrimp ovarian extract. The antiserum was then used to precipitate vitellogenin from female shrimp haemolymph and this precipitate was used to immunize the second set of mice. This antiserum made against vitellogenin was used to assess vitellogenin levels by competitive ELISA. Haemolymph vitellogenin levels of female shrimps were directly related to their ovarian developmental stages. The vitellogenin levels were low (0.193+-0.167 mg/ml) during the resting stage, high (2.3-2.6 mg/ml) during the developing stages, and decreased (0.708+-0.508 mg/ml) during the ripen stage. During ovarian degeneration, the vitellogenin levels were unusually high (15-25 mg/ml). Application of eyestalk extract to eye-ablated shrimps induced elevation of the haemolymph vitellogenin to very high levels which may result from degeneration of the ovary.
