Effects of light and humidity in in vitro conditioning of Asparagus plantlet prior to transplanting

Asparagus shoot were cultured on a MS medium supplemented 0.04 mg/l IBA and 60 g/l sucrose under white light at photosynthetic photon flux (PPF) of 28, 37 and 44 micro mol. 1/square m/s, blue light at PPF of 21, 29 and 40 micro mol/square m/s and red light at PPF of 12, 16 and 25 micro mol/square m/s of red light. After transplanting for 6 weeks, all of them were survived. The tallest plantlets and the longest root length were found under white light at PPF 37 micro mol/square m/s and 28 micro mol/square m/s, respectively. The number of shoot/clump and the whole plant fresh weight. Asparagus shoot were cultured on a MS medium supplemented 0.04 mg/l IBA and 60 g/l sucrose at 32, 56 and 90 percent relative humidity in the culture vessel that had a hole of 0, 0.5 and 1.0 cm diameter on its closure. After transplanting for 6 weeks, the tallest plantlets of 22.83 cm were found at 32 percent relative humidity in the culture vessel that no hole on its closure. The highest whole fresh weight of 4.81 g were found at 32 percent relative humidity in the culture vessel that had a 0.5 cm diameter on its closure. The root length and the number of shoot/clump and the whole plant fresh weight of plantlets from different air humidity condition.
