Clonal propogation of mangosteen (Garcinia mangostana Linn.) through friable callus

Clonal propagation of mangosteen (Garcinia mangostana Linn.) was conducted using friable callus (5x5 mm) induced from young leaves on MS (Murashige and Skoog, 1962) medium with 3 percent sucrose, 500 mg/l PVP (polyvinylpyrrolidone), 0.17 percent phytagel, 0.5 mg/l BA (benzlyadenine) and various concentrations (1-50 mg/l) of 2,4-D (2,4 dichlorophenoxyacetic acid) in the dark for 4 weeks. The results showed that 1 mg/l 2,4-D gave the highest increase in fresh weight of callus at 420 mg, size 6.7 square m and average proliferation rate 68.06 percent. The callus was friable, yellowish cream to brown in color. When the concentration of 2,4-D was increased to 2.5 to 50 mg/l, the fresh weight and size of callus decreased drastically and the callus turned to brown color. When the callus was subcultured onto meristematic nodular callus induction medium (MS with 3 percent sucrose, 500 mg/l PVP, 0.5 mg/l BA and 0.5 mg/l TDZ (thidiazuron) and maintained under light for 8 weeks, the highest nodular callus of 22.22 percent and the number of nodules of 1.42 per callus were obtained.
