Comparative study of simple methods for the diagnosis of yellow-head baculovirus in the black tiger shrimp (Penaeus monodon)

Six current methods were used to detect the yellow-head baculovirus (YBV) infection in black tiger shrimp (Penaeus monodon). They were : total hemocyte count, wet mount of the gills, copper content in hemolymph, Giemsa staining of hemocytes, hematoxylin staining of the gills and scanning electron microscopic (SEM) studies of hemocyte surface. Wet mount of the gills and hematoxylin staining of the gills had poor results in detecting the early stages of yellow-head baculovirus infection. The techniques of copper content of hemolymph, Giemsa staining of hemocytes and total hemocyte count showed promising result which could be useful for YBV diagnosis. But more baseline data are needed to standardize these methods.
