Detection of DNA polymorphisms in Thai aromatic rice using RAPD-PCR assay

DNA polymorphism of nine aromatic rice varieties (Oryza sativa L.) was examined by using the random amplified polymorphic DNA (RAPD) technique. Six random primers were used to amplify DNA fragments. The amplifield products were separated on agarose gel and banding patterns were observed. A total of 42 RAPD bands were generated by the six primers. Of these, 23 (54.8 percent) bands were polymorphic. Average number of total amplified bands for each primer was 8.4. By comparing the banding patterns, the proportion of common bands between the nine varieties was calculated and a dendrogen showing genetic relationships was constructed. The level of genetic variation estimated by squared euclidean dissimilarity coefficient, ranging from 0.01 to 0.64, between the nine varieties may be explained by the broad genetic base existing in amoung genomes of these nine varieties.
