Study on interspecific hybridization of Brassica spp. through in vitro culture

Imbibed seed of B. campestris and B. juncea were vernalized for one month and juvenile seedlings of B. oleracea were vernalized for one half month at 5 deg C under 16 light condition, in order to induce flowering. All plants were transferred into the growth cabinet in which controlled 23-25 deg C, continuous light condition. After 20 days cross pollination among species were made, the hybrid capsules were collected and embryos were rescued in the modified MS medium supplemented with casein hydrolysate, BA (0.1 mg/l) and NAA (0.1 mg/l). The embryo of hybrids between B. juncea and B. oleracea; B. campestris and B. juncea; B. campestris and B. oleracea were successfully regenerated to plantlets after 30 days in vitro culture.
