Screening of microorganisms for amylase, protease and lipase production

Isolation of microorganisms producing amylase, protease and lipase at 37 deg C and 55 deg C was carried out by selection of microorganisms grew on selective medium as starch agar, skim milk agar and tributyrin agar. 104 and 34 isolates of amylase producers, 63 and 47 isolates of protease producers and 82 and 47 isolates of lipase producers were screened at 37 deg C and 55 deg C respectively. The microorganisms producing clear zone diameter more than 1 cm. were selected and confirmed for amylase, protease and lipase activity in liquid medium by quantitative assay. Among them, A and JA8 showed high amylase activity at 37 deg C while BS and TR showed at 55 deg C, CHB9 and JB33 showed high protease activity at 37 deg C as well as 1.4 SA and C2 at 55 deg C, AUA2 and M18 showed high lipase activity at 37 deg C and also TR and BS at 55 deg C. The microorganisms were all identified as genus Bacillus.
