Seeds of rice (Oryza sativa L.) cultivar Khao Kawk Mali 105 were cultured on LS (Linsmaier and Skoog, 1965) medium supplemented with 2 mg/l 2,4-D and 0.5 mg/l kinetin. Callus was initiated from these seeds within 7 days. The cultured seeds were harvested at various times, fixed, sectioned, stained and examined micro-scopically. The histological origin of the callus revealed that callus formation occurred from cells with dense cytoplasm in vascular tissue of mesocotyl and epithelium tissue of scutellum.
