Repetitive nuclear DNA sequence of DNA probe for intraspecies analysis of Apis cerana

Studies of genetic variation among Apis cerana populations has been performed by Uthaisang (1994) using Restriction Fragment Length Polymorphisms (RFLPs) analysis. Six different RFLPs patterns were obtained from repetitive DNA probe no.99 which was a pUC118 recombinant plasmid comprising 2.3 kb chromosomal DNA fragment of A. cerana. The present study aims to determine the repetitive sequence of the DNA probe no.99 which could be a useful tool to identify and characterize genetic variation in A. cerana. The 2.3 kb insert fragment from DNA probe no.99 was subcloned as sequenced. Two subclones with 0.94 kb and 1.38 kb of inserted fragments were constructed. Sequencing results showed that inserted fragments of both subclones have high content of A and T. In the 0.94 kb inserted fragment, the sequence (TTATAAAAATAAACTTT) was observed and may be a minisatellite DNA. Study will be further conducted to confirm this result. Minisatelite DNA pattern was no found in the 1.38 kb insert fragment.
