Molecular profiling of rice cultivars using RAPD and AFLP marker systems [Oryza sativa L.; India; Randomly Amplified Polymorphic DNA; Amplified Fragment Length Polymorphism]

A total of 18 accessions of Oryza sativa subspecies indica were subject to molecular marker analysis involving a set of 14 random decamer primers and six AFLP primer combinations to produce 145 and 291 RAPD and AFLP markers respectively. The number of polymorphic markers ranged from 7 to 13 for RAPD markers and from 9 to 33 for AFLP markers. Both the marker systems grouped the accessions into two groups of traditional and evolved genotypes from IRRI. Though the level of polymorphism for AFLP makers was low (35.40%) when compared to RAPD markers (88.97%), the grouping based on AFLP markers remained more accurate than the RAPD markers based grouping. These unmapped multiloci marker systems (RAPD and AFLP) are expected to be useful in assessing the extent of genetic diversity in other crops where mapped markers are not available

Il numero di marcatori polimorfici variava da 7 a 13 per i marcatori RAPD e da 9 a 33 per i marcatori AFLP. Ambedue i sistemi di marcatori hanno raggruppato le accessioni in due gruppi di genotipi tradizionali ed evoluti provenienti dall´IRRI. Sebbene il livello di polimorfismo dei marcatori AFLP fosse basso (35,40%) rispetto ai marcatori RAPD (88,97%), il raggruppamento basato sui marcatori AFLP rimaneva più accurato di quello basato sui marcatori RAPD. Si può ritenere che questi sistemi di marker multiloci non mappati (RAPD e AFLP) siano utili per accertare il livello di diversità genetica in altre specie coltivate per cui non sono disponibili marcatori mappati.]
