Comparative performance of androgenesis and maize-mediated systems of polyhaploid induction in wheat and triticale [Triticum aestivum L.; x Triticosecale Witt.]

The investigation was undertaken to evaluate the comparative efficiency of anther culture and maize-mediated (wheat x maize) systems of haploid induction in wheat (Triticum aestivum L.) and triticale (x Triticosecale) genotypes. Ten genotypes of each were subjected to anther culture and were also simultaneously crossed with the Madgran Local genotype of maize. Considerable genetic variability was observed for all the component traits of haploid induction through the two techniques. The haploid formation frequencies through wheat x maize system were significantly higher, both in wheat (73.48%) and triticale (45.71%) genotypes as compared to the calli induction frequencies through anther culture (8.66 and 2.68%, respectively). Two wheat and four triticale genotypes did not respond to anther culture, whereas all wheat and triticale genotypes formed a sufficient number of haploid embryos through the wheat x maize system. The haploid plantlet regeneration frequencies were also significantly higher through the latter technique in wheat (24.46%) and triticale (27.82%) as compared to anther culture (14.80 and 11.77%, respectively). The overall success rate of obtaining green haploid plantlets per 100 florets pollinated/anthers cultured was also significantly higher through the wheat x maize technique (4.39% in wheat and 1.70% in triticale) in comparison to androgenesis, where the frequencies were only 1.83 and 0.44%, respectively. [E´ stata confrontata l´efficienza della coltura di antere e di sistemi mediati dal mais (frumento x mais) nell´induzione di aploidi in genotipi di frumento (Triticum aestivum L.) e di triticale (x Triticosecale). Dieci genotipi delle due specie sono stati sottoposti a coltura di antere e contemporaneamente incrociati con il genotipo di mais Madgran Local. E´ stata osservata un´ampia variabilità genetica per tutti i caratteri interessanti l´induzione di aploidi nell´ambito delle due tecniche. Le frequenze di formazione di aploidi con il sistema frumento x mais erano significativamente maggiori, nei genotipi sia di frumento (73,48%), sia di triticale (45,71%), rispetto all´induzione di calli da coltura di antere (8,66 e 2,68%, rispettivamente). Due genotipi di frumento e quattro di triticale non hanno risposto alla coltura di antere, mentre tutti i genotipi di frumento e triticale hanno formato un numero sufficiente di embrioni aploidi con il sistema frumento x mais. Le frequenze di rigenerazione di piantine aploidi sono risultate significativamente maggiori con quest´ultima tecnica nel frumento (24,46%) e nel triticale (27,82%) rispetto alla coltura di antere (14,80 e 11,77%, rispettivamente). Il successo complessivo nell´ottenimento di piantine aploidi verdi per 100 fiori impollinati/antere coltivate era pure significativamente maggiore con la tecnica frumento x mais (4,39% nel frumento e 11,77% nel triticale) in confronto all´androgenesi, dove le frequenze sono state, rispettivamente, solo dell´1,83 e dello 0,44%. Nel caso della coltura di antere, limiti quali alta specificità genotipica, albinismo e bassi tassi di rigenerazione riducevano l´efficienza della tecnica.]
