Development of disposable aspirating devices for collection of pig follicular fluid and oocytes

The present study was carried out to develop disposable devices which permited effective collection of porcine follicular fluid and cumulus-oocyte complexes (COCs). The constructions of these devices are shown in Figs. 1 and 3. Collection of follicular fluid When different suction pressures (50-400 mmHg) were applied to pFF collecting chamber (Fig 1), the amounts of follicle fluid collected for 10 min were significantly larger as compared to the conventional method using syringe (Fig 4). When oocytes were cultured in the mediums containing pFF collected by different methods, the abilities of maturation and development of oocytes were not influenced appreciably (Table 1). Collection of cumulus-oocyte complexs (COCs) Influences of suction pressures (10-80 mmHg) on the efficacy of collecting COCs and these qualities were tested. The suction pressure of 40 and 80 mmHg increased the number of COCs collected for 1 min significantly compared with that of 10 mmHg and the conventional method using syringe (Fig 5). The quality (Rank) of COCs was not influenced by these collecting methods (Fig 6). The abilities of maturation and development of oocytes (Rank A) after culture were not influenced by these collecting methods. The results of present study indicate that efficacy of collecting pFF and COCs is remarkably improved by the use of disposable aspirating devices as compared to the conventional method using syringe without influencing the qualities of pFF and COCs.
