Mutagenicity study of Tawa-tawa (Euphorbia hirta) plant decoction

The mutagenicity study of Tawa-tawa(Euphorbia hirta) plant decoction was carried out to assess if the plant can cause changes in genetic material in individuals which can be passed from one generation to another. The investigation utilized Ames Test which used Salmonella typhenurium TA 98 as test strain. The strain was tested with 20%, 40%, 60%, 80% and 100% Tawa-tawa plant decoction corresponding to cocentrations of 193.1, 286.3, 579, 772.6, and 965.7 mg/ml, respectively. Acridene orange was used at a concentration of 2mg/ml as positive control, and distilled water as the negative cotrol. There were three trials with three replicates per trial. The mutagenic activity of the plant was evaluated using a "modified two-fold increase". This was done by comparing the number of revertants observed in the treated cultures to those in untreated cultures. Mutagenicity was assessed if the mean number of colonies exhibited by Tawa-tawa at different concentrations were twice the number of visible colonies exhibited against distilled water as negative control. Based on the data gathered, the mean number of visible colonies in plates treated with 20%, 40%, 60%, 80% and 100% decoction were 113, 140, 161, 229, and 370 CFU/ml, respectively. As expected the mean number of visible colonies in plates treated with acridine orange was 265. On the other hand, the mean number of visible colonies in plates treated with distilled water as negative control was 79 CFU/ml. It was shown that the number of visible colonies in plates treated with 60%, 80%, and 100% were twice the spontaneous level.
