Differentiation of local Fusarium isolates using RAPD-PCR

Fusarium fungi, responsible for serious diseases, are a grave threat to arable crops. The genus contains many species and physiological races, which makes unambiguous identification difficult if traditional methods based on morphological characteristics are applied. Therefore, the present laboratory experiment has been conducted as an attempt to differentiate some isolates of the Fusarium genus (F. avenaceum, F. culmorum, F. oxysporum and F. poae) based on genetic differences by applying molecular method. The polymerase chain reaction (PCR) and its modifications was the basic research method. The RAPD-PCR method was used for differentiation between selected fungi. The results showed that 7 out of the 9 short 10-mer RAPD primers used in the assays yielded positive results and enabled differentiation between the four isolates of Fusarium
