Non-starch polysaccharides of taro (Colocasia esculenta)

This study aimed at providing more detailed information about the extraction, purification and composition analysis of non-starch polysaccharides (NSPs) of taro. An improved extraction scheme for taro NSPs was developed following the sequence: taro-taro flour-CWX**(2)-taro starch-3NX**(2)-residues. The purification of taro NSPs included dispersion and hydrolysis of starch, followed by removal of starch hydrolysate and unconjugated protein. Ion exchange chromatography of purified taro NSP showed that purified taro CWX and 3NX could be further fractionated into neutral or acidic NSPs. The analysis of fractions of purified taro CWX confirmed that CWXI or CWXII were free of starch, and combined with some uniform protein, and that CWXII represented an acidic NSP fraction. The analysis of monosaccharides in the hydrolysates of fractions of purified taro NSPs by GC-MS and HPAEC-Pad showed mainly galactose and arabinose. Small quantities of glucose and mannose were found, but xylose and rhamnose were not detected in the fractions of purified taro CWX.
