Methods of analysis of cholesterol in animal fats

In the case of analysis of total cholesterol it is necessary its liberating from esters by pre-treatment with fatty acids.Three methods for pre-treatment were considered: saponification following AOAC protocol, metylation using boron trifluorid and methylation using tetramethyl ammonium hydroxide (TMAH). From bobliographic review it was showed that the methylation using TMAH could not assure the necessary precision, when both other methods give the comparative results.
