Mitochondrial mutations in maize [Zea mays L.]

English. Maize is an ideal organism in which to study mitochondrial DNA (mtDNA) mutations. Three different types of male-fertile (NA, NB and RU) and three types of male-sterile (C, T and S) mitochondrial genotypes have been characterized in maize. Reversions of cytoplasmic male sterility (CMS) to fertility and mutations resulting in abnormal growth, termed nonchromosomal stripe (NCS) in maize, are readily detected. Male-fertile revertants have been observed in field-grown CMS-S plants as well as in plants regenerated from CMS-T and CMS-S tissue culture. NCS mutants arising in CMS-T, CMS-S sterile and CMS-S revertant cytoplasms have also been characterized. The nuclear genotype-cytoplasm combination can be manipulated in order to study the effects of nuclear genes on the frequency and type of mitochondrial mutations. Revertants of CMS to fertility, as well as abnormal growth mutants, were observed by early maize geneticists, but such mutations required advances in technology to determine that they resulted from changes in mtDNA. It is now known that specific mtDNA deletions initiated by rare aberrant recombination across short repeat regions are most often the cause of these mutations

Nel mais sono stati caratterizzati tre tipi diversi di genotipi mitocondriali maschio-fertili (NA, NB ed RU) e tre tipi di maschiosterili (C, T e S). Le reversioni della sterilità citoplasmatica maschile (CMS) alla fertilità e le mutazioni che si traducono in una crescita anormale, denominata nonchromosomal stripe (NCS) nel mais, sono riscontrabili prontamente. Genotipi che ritornano a maschio-fertili sono stati osservati in piante CMS-S in campo, così come in piante rigenerate da colture di tessuti CMS-T e CMS-S. Sono stati pure caratterizzati mutanti NCS che si originavano in citoplasmi CMS-T, CMS-S sterili e CMS-S che ritornavano. La combinazione genotipo nucleare-citoplasma può essere manipolata per studiare gli effetti dei geni nucleari sulla frequenza e sul tipo di mutazioni mitocondriali. Piante di CMS che ritornavano alla fertilità, così come mutanti a crescita anormale, sono stati osservati dai primi genetisti del mais, ma tali mutazioni richiedevano progressi tecnologici per determinare che derivavano da cambiamenti del mtDNA.
