Effects of colchicine added to the induction medium on the regeneration ability of calli in the wheat anther culture

The aim of this study was to investigate effects of colchicine, added to the induction nutrient medium on the regeneration ability of calli in the wheat (Triticum aestivum L.) anther culture. F2 generation of 14 different hybrid combination of wheat (Triticum aestivum L.) were used. From each combination were isolated a total of 1200 anthers. A half of them was exposed to the colchicine effect for three days on an modified Potato-2 induction medium with 300 mg/l colchicine, while the other half was grown in the v colchicine-free medium. The combination BanksxF53-70 was the most susceptible to the presence of colchicine in the induction medium. In this combination, whole regeneration ability of calli was 40.9% and 15.0% on medium without and with colchicine, respectively. A total of 88 green and 65 albino plants was regenerated on the colchicine-free medium while 35 green and 35 albino plants were developed on the colchicine-supplemented medium.
