Haploid regeneration aptitude of maize (Zea mays L.) lines of various origin and of their hybrids

One maize line with good haploid regeneration aptitude and one recalcitrant in this respect, together with various hybrid combinations of these, were used to develop new doubled haploid (DH) lines. A line originating from an exotic Chinese stock was crossed with a recalcitrant Martonvasar line, used as a parental component in several commercial hybrids, to determine the anther response, callus induction and plant regeneration of the reciprocal hybrids and the fertility of the adult plants. The same tests were also made on previously developed DH lines of these reciprocal hybrids and on DH lines obtained after a single backcross with the recalcitrant parent. The results indicated that haploid regeneration aptitude was primarily an inherited trait in the experimental material, with a heterosis effect in the F1 progeny. The hybrids exhibited no statistically significant reciprocal effect. The anther response, callus induction and plant regeneration of the DH lines developed from the hybrids and the fertility of regenerated plants were comparable to the initial combinations even when the DH line originated from a hybrid developed by backcrossing with the recalcitrant line. Data are also reported on the anther response and haploid regeneration frequency of genotypes known to have excellent tissue culturing aptitude from immature zygotic embryos. The present results may contribute to the development of an optimum gene ratio where, in addition to maintaining haploid regeneration aptitude, the DH lines also possess important agronomic traits, which are then reliably inherited

[Una linea di mais con buona attitudine alla rigenerazione aploide e una recalcitrante per questo aspetto, unitamente a varie combinazioni ibride di queste, sono state utilizzate per sviluppare nuove linee doppio-aploidi (DH). Una linea originata da un materiale esotico cinese è stata incrociata con una linea recalcitrante Martonvasar, utilizzata come componente parentale in numerosi ibridi commerciali, per determinare la risposta delle antere, l'induzione del callo e la rigenerazione della pianta degli ibridi reciproci e la fertilità delle piante adulte. Gli stessi test sono stati anche effettuati su linee DH di questi ibridi reciproci sviluppate precedentemente e su linee DH ottenute da un singolo reincrocio con il parentale recalcitrante. I risultati hanno indicato che l'attitudine alla rigenerazione aploide era primariamente un carattere ereditato nel materiale oggetto di esperimento, con un effetto eterotico nella progenie F1. Gli ibridi non hanno mostrato alcun effetto reciproco significativo. La risposta delle antere, l'induzione del callo e la rigenerazione delle piante delle linee DH sviluppate dagli ibridi e la fertilità delle piante rigenerate sono risultate confrontabili con le combinazioni iniziali anche quando la linea DH traeva origine da un ibrido sviluppato mediante reincrocio con la linea recalcitrante. Sono pure riportati dati sulla frequenza di risposta delle antere e di rigenerazione aploide di genotipi noti per la buona attitudine alla coltura di tessuti da embrioni zigotici immaturi. Questi risultati possono contribuire allo sviluppo di un rapporto genico ottimale in cui, oltre al mantenimento dell'attitudine alla rigenerazione aploide, le linee DH posseggono pure caratteri agronomici importanti, che sono quindi ereditati attendibilmente.]
